MOTS-c was identified through analysis of short open reading frames within mitochondrial 12S ribosomal RNA, establishing it as one of the mitochondrial-derived peptides.1 In published preclinical models, research has examined its role in AMP-activated protein kinase (AMPK) signaling and the folate–methionine one-carbon cycle, positioning the peptide within mitochondrial signaling research and cellular metabolism research.1,2 Subsequent work reported that MOTS-c can translocate to the nucleus under conditions of metabolic stress, where it has been investigated for its role in the regulation of nuclear gene expression and stress-adaptive transcriptional pathways.3 Additional studies have characterized MOTS-c as an exercise-associated, mitochondrial-encoded factor examined in the context of skeletal-muscle homeostasis research and age-associated physiological change in rodent model systems.4 Collectively, this literature frames MOTS-c as a tool compound for investigating mitochondria-to-nucleus retrograde signaling and metabolic-homeostasis research pathways.
